The Designare Z390 with built-in Titan Ridge is looking cheaper and cheaper by the minute!! :)
the card and a new PSU are less that the Z370 Designaire and the time of a tear down and rebuild also add to the deficit equation. sheeeeesh!
 
The adapters are inexpensive, will they work?
Molex should be avoided as much as possible. The SATA to PCIe 6-pin would be the preferred option.

Well, this won't work at all. Since it needs a USB port, the only way that would work is to lose the front case USB ports and that would also lose both card readers

I use this internal USB 2 splitter that works perfectly. It provides four USB 2 connections through a hub built into the little printed circuit board (PCB).

Saw that. The weird thing is that I have a powered internal USB hub but the Bluetooth USB (part of the wifi PCIe adapter) will not work when plugged into the internal 9 pin plug. (https://www.amazon.com/gp/product/B01IFGFTJ2/?tag=tonymacx86com-20)

Ended up buying this and a pair of SATA>PCIe 6 plugs:

One thing that I am concerned about with this add-in USB card, will that not change the number of ports in the system?

No, it's on a separate controller so it won't be counted towards the chipset USB ports.

Oops. Thought you were talking about a PCI-e USB card... No. The thing you are looking at is like a hub. It doesn't add ports.
