Okay, I have done what. But it does not solve the issue.
If I start the system via the USB drive, choose there instead of "USB" the "Lion_SSD", it will enter Lion on the SSD.
Audio and everything is working as expected. So only the Bootloader "MultiBeast" is somehow missing.

Problem found and solved. Sierra does not stable support the Sapphire Radeon HD 6850.
Have removed it from the Motherboard and did inject the I7 GPU HD 3000 via Clover (=config.plist) + installed the MultiBeast-Kext for HD 3000. Connection via Onboard Motherboard HDMI port to the monitor.
