Error: No boot disk has been detected, no audio, no wifi, gtx 550ti

I have a Hp Envy 700-074, Intel i5 4430, 12GB Ram, Nvidia GTX 550ti (not installed yet)

I am getting a "error: no boot disk has been detected or the disk has failed" after I perform a reboot of my machine.
To get back into my machine I will need to press escape at boot up and choose the usb and the machine will boot just fine.

I have got Sierra installed and running, I have internet via ethernet.

I am having problems with post installation also getting the proper audio drivers, wifi and I have a nvidia gtx 550ti video card I would like to install and working properly (I did install it, and got a smaller screen size with lots screen flicker, so removed it and went back to onboard gpu which works)

I tried the multibeast but Im lost deciphering which codec and other kexts to install to get a stable machine.

Any help will be greatly appreciated.
