You need to prevent the AMD GPU from getting initialized during boot phase, using iGPU=Primary is the most common approach to do this.

Installing a 2nd "helper GPU" can do the same for you. Depending on your mainboard's UEFI implementation it could also be possible to unplug your monitor during boot phase.
It's also possible to modify the VBIOS to prevent it from getting utilized during boot phase. I wouldn't recommend to do this unless your card has a dual BIOS, because it could easily result in a brick.

I think is the only way to boot and have GPU running with acceleration : but in my system repport I have HD 4400 set.

Have you tried to use R9 380 with the El Capitan?

No : i read post here that in El Capitan it may be also difficult, and I have install El Capitan in other builds : I do not like some spec .

Yosemite is quite good ( for exemple disk utility) and may be I also return on Yosemite :

It's for me "rediculous" to have every time to unplug and replug display cable : it'not what we could name plug and play ...

- Yosemite yes work OOB
- El Capitan, Sierra & High Sierra Beta with helper card or IGP as primary in bios
