macOS 10.13.3 Supplemental Update

That advice must have been given long time ago. Since Sierra, MultiBeast has always used /Library/Extensions/ folder which Apple started recommending with 10.12 Sierra.
I ran sudo kextcache -I / (small "I" I cannot stop auto correct changing it to cap I) see attached, and found many third party kexts in S/L/E. The question is, should they be removed, simply by deleting/dragging to trash and should they all be copied to "kexts/other" in clover folder?

If you installed macOS according to the tonymacx86 installation guide using UniBeast and MultiBeast, then, no, you do not need to remove/move any kext from /S/L/E.

I did and I made some changes since then these concern me as well as the USB inject all kext being in S/L/E

what would be the circumstances that one would need this?

If you want graphics to work as similarly as it does on a real Mac as possible. It's possible that your graphics card may work without the rename, but GFX0 is where macOS expects the video card to be. It's up to you if you want to do the rename or not. Personally, I always use it.

And GFX0 to IGPU? This is what I have in patches.
 
As the rename implies, it's for the IGPU.
Pretty much what I figured but sometimes I need to ask the foolish question with the obvious answer . Is it needed when using a graphics card?
